Mycobacteria 유전체 분석 #

Mycobacterium tuberculosis complex는 결핵의 원인균으로 전 세계적으로 연간 천만 명의 신환자가 발생하고 있으며, 국내의 경우 해마다 20,000여명 이상의 신환자가 보고되고 있다. 여전히 진단과 치료에 어려움을 겪고 있으며, 특히, 기존의 결핵균 유전형 검사법은 변별력, 재현성, 검사자의 생물안전 문제 등으로 보다 변별력 높은 검사법이 요구되고 있다. 분석기술의 발전에 따라 최근에는 전장유전체염기서열(Whole genome sequencing, WGS) 분석법이 대중화되었으며 결핵균에서 WGS를 활용하는 사례가 증가하고 있다. 결핵 유사 질병을 일으킬 수 있는 subtypes 들의 유전체 변이는 0.01%이기 때문에 유전적 차이에 근거한 미세 변이를 찾아내는 것이 중요하다. 이러한 변이는 주로 면역 탈출(immune evasion), 항원 변이(antigenic variation) 및 병독성(virulence)과 관련되어 있으며, 또한 결핵균 치료에 중요한 1차 항생제에 대한 약제 내성을 유도할 수 있는 것으로도 보고되었다. 따라서 이러한 미세 변이의 정확한 규명은 보다 효과적인 진단법 및 치료 전략을 개발하는 데 중요하다. 현재 유전체 수준에서 다양한 데이터가 쌓이고 있으며, 아래의 Mycobacteria 전문 데이터베이스를 통해 미세변이를 분석할 수 있다.
